Abstract

Gerbera (Gerbera jamesonii) merupakan salah satu komoditas bunga potong yang banyak diperdagangkan, namun perbanyakannya secara konvensional menghasilkan benih dalam jumlah terbatas sehingga perbanyakan in vitro menjadi alternatif untuk mendukung penyediaan bibit secara massal. Salah satu tahap kritis dalam kultur jaringan adalah sterilisasi eksplan, karena tingkat kontaminasi yang tinggi sering menjadi kendala utama keberhasilan inisiasi kultur. Penelitian ini bertujuan untuk menganalisis pengaruh variasi konsentrasi alkohol (20%, 30%, dan 40%) terhadap tingkat keberhasilan sterilisasi eksplan batang Gerbera secara in vitro, serta menentukan konsentrasi alkohol optimal yang mampu meminimalkan kontaminasi tanpa menyebabkan kerusakan jaringan (browning). Penelitian dilaksanakan di Laboratorium Kultur Jaringan Universitas Labuhanbatu pada bulan Mei–Juli 2026, menggunakan Rancangan Acak Lengkap (RAL) satu faktor dengan tiga taraf perlakuan dan enam ulangan, sehingga diperoleh 18 unit percobaan. Data dianalisis menggunakan Analisis Sidik Ragam (ANOVA) pada taraf 5%, dilanjutkan dengan Uji Jarak Berganda Duncan (DMRT) apabila terdapat pengaruh nyata. Hasil penelitian menunjukkan bahwa perbedaan konsentrasi alkohol tidak berpengaruh nyata terhadap persentase sterilitas eksplan (F hitung 0,20 < F tabel 5% 3,68). Persentase sterilitas tertinggi diperoleh pada perlakuan alkohol 20% dan 30% (masing-masing 67%), sedangkan alkohol 40% menghasilkan sterilitas terendah (50%). Seluruh kontaminasi yang teramati (100%) disebabkan oleh bakteri, tanpa ditemukan kontaminasi jamur, yang mengindikasikan sumber kontaminasi berasal dari bakteri endofit di dalam jaringan eksplan, bukan dari kegagalan sterilisasi permukaan. Berdasarkan pertimbangan efisiensi dan risiko fitotoksisitas, konsentrasi alkohol 20% direkomendasikan sebagai batas sterilisasi optimal untuk eksplan batang Gerbera. Kata Kunci : Gerbera Jamesonii; Sterilisasi Eksplan; Konsentrasi Alkohol; Kultur Jaringan; In Vitro; Kontaminasi Bakteri ================================================================================================= Gerbera (Gerbera jamesonii) is a widely traded cut-flower commodity, but conventional propagation yields a limited number of seedlings, making in vitro propagation an alternative for mass seedling production. One of the critical stages in tissue culture is explant sterilization, as a high contamination rate is often the main obstacle to successful culture initiation. This study aimed to analyze the effect of varying alcohol concentrations (20%, 30%, and 40%) on the sterilization success rate of Gerbera stem explants in vitro, as well as to determine the optimal alcohol concentration that minimizes contamination without causing tissue damage (browning). The research was conducted at the Tissue Culture Laboratory of Universitas Labuhanbatu from May to July 2026, using a one-factor Completely Randomized Design (CRD) with three treatment levels and six replications, resulting in 18 experimental units. Data were analyzed using Analysis of Variance (ANOVA) at the 5% level, followed by Duncan's Multiple Range Test (DMRT) when significant effects were found. The results showed that differences in alcohol concentration had no significant effect on explant sterility percentage (F calculated 0.20 < F table 5% 3.68). The highest sterility percentages were obtained with the 20% and 30% alcohol treatments (67% each), while the 40% alcohol treatment produced the lowest sterility (50%). All observed contamination (100%) was caused by bacteria, with no fungal contamination found, indicating that the source of contamination originated from endophytic bacteria within the explant tissue rather than from surface sterilization failure. Based on efficiency and phytotoxicity risk considerations, a 20% alcohol concentration is recommended as the optimal sterilization limit for Gerbera stem explants.
