Abstract

Sterilisasi eksplan merupakan tahap krusial dalam kultur jaringan tanaman, karena eksplan yang diambil dari lingkungan alami membawa berbagai mikroorganisme permukaan yang berpotensi menjadi sumber kontaminasi pada media kultur. Penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi alkohol terhadap persentase eksplan steril dan kontaminasi, serta mengetahui kecenderungan waktu muncul kontaminasi dan jenis mikroorganisme kontaminan pada eksplan biji Antigonon leptopus secara in vitro. Penelitian dilaksanakan menggunakan Rancangan Acak Lengkap (RAL) satu faktor dengan tiga taraf perlakuan konsentrasi alkohol, yaitu 20% (P1), 30% (P2), dan 40% (P3), masing masing diulang sebanyak 6 kali. Eksplan disterilisasi melalui tahapan bertingkat meliputi pencucian dengan sabun, perendaman betadine dan Dithane, sterilisasi alkohol sesuai perlakuan, serta perendaman bayclin bertingkat (15% dan 20%) di dalam Laminar Air Flow (LAF). Hasil penelitian menunjukkan bahwa perlakuan konsentrasi alkohol tidak berpengaruh nyata terhadap persentase eksplan steril (F hitung 0,682 < F-tabel 3,68 pada taraf uji 5%), dengan ketiga perlakuan berada pada notasi DMRT yang sama. Meskipun demikian, P3 (40%) menunjukkan kecenderungan hasil terbaik dengan persentase eksplan steril tertinggi (84%) dan kontaminasi terendah (16%), sedangkan P2 (30%) menunjukkan performa paling kurang optimal (kontaminasi 50%). Rata-rata waktu muncul kontaminasi berkisar antara 6-7 hari setelah tanam (HST), dengan jenis kontaminan yang didominasi oleh jamur (33% pada P1 dan P2, 16% pada P3), sedangkan kontaminasi bakteri hanya ditemukan pada P2 (17%). Keberhasilan sterilisasi pada penelitian ini lebih dipengaruhi oleh akumulasi tahapan sterilisasi bertingkat dibandingkan oleh konsentrasi alkohol secara tunggal. Kata Kunci : Antigonon Leptopus, Sterilisasi Eksplan, Alkohol, Kultur Jaringan, Kontaminasi =============================================================================================== Explant sterilization is a crucial stage in plant tissue culture, as explants collected from natural environments carry various surface microorganisms that may become sources of contamination on culture media. This study aimed to determine the effect of alcohol concentration variation on the percentage of sterile and contaminated explants, as well as to identify the tendency of contamination onset time and the type of contaminating microorganisms on Antigonon leptopus seed explants cultured in vitro. The study was conducted using a Completely Randomized Design (CRD) with one factor consisting of three alcohol concentration treatments, namely 20% (P1), 30% (P2), and 40% (P3), each replicated six times. Explants were sterilized through a multi-stage procedure comprising washing with dish soap, immersion in betadine and Dithane solutions, sterilization with alcohol according to treatment, and graded bleach (bayclin) immersion (15% and 20%) inside a Laminar Air Flow (LAF) cabinet. The results showed that alcohol concentration treatment had no significant effect on the percentage of sterile explants (F-calculated 0.682 < F-table 3.68 at the 5% significance level), with all three treatments falling under the same DMRT notation. Nevertheless, P3 (40%) exhibited the best tendency, with the highest percentage of sterile explants (84%) and the lowest contamination rate (16%), whereas P2 (30%) showed the least optimal performance (50% contamination). The average contamination onset time ranged between 6-7 days after planting (DAP), with fungi being the dominant contaminant (33% in P1 and P2, 16% in P3), while bacterial contamination was only found in P2 (17%). The success of sterilization in this study was more strongly influenced by the accumulation of multi-stage sterilization steps than by the alcohol concentration alone.
