Abstract

Optimalisasi Sterilisasi Eksplan Tunas Kopi Robusta (Coffea canephora) untuk Penanaman In Vitro. Dibimbing oleh Kamsia Dorliana Sitanggang dan Badrul Ainy Dalimunthe. Penelitian ini bertujuan mengevaluasi konsentrasi alkohol 40%, 50%, dan 60% pada sterilisasi permukaan eksplan tunas kopi robusta. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Universitas Labuhanbatu pada Mei-Juli 2026. Penanaman dilakukan pada 1 Juli 2026 dan pengamatan berlangsung sampai 8 hari setelah tanam. Penelitian menggunakan Rancangan Acak Lengkap satu faktor dengan tiga taraf perlakuan dan enam ulangan, sehingga terdapat 18 unit percobaan. Alkohol 40% menghasilkan 33,33% eksplan bebas kontaminasi, sedangkan alkohol 50% dan 60% masing-masing menghasilkan 50,00%. Rata-rata waktu muncul kontaminasi berturut-turut adalah 7; 7; dan 8 HST. Kontaminasi didominasi jamur. Analisis ragam terhadap data berkode 100 untuk bebas kontaminasi dan 0 untuk terkontaminasi menghasilkan F-hitung 0,192 dengan p = 0,827. DMRT 5% memberikan notasi yang sama pada semua perlakuan. Uji eksak Fisher-Freeman-Halton juga tidak menunjukkan hubungan yang signifikan antara konsentrasi alkohol dan status kultur (p = 1,000). Secara deskriptif, alkohol 60% menunjukkan kecenderungan paling efektif karena menghasilkan persentase bebas kontaminasi tertinggi bersama alkohol 50% dan waktu muncul kontaminasi paling lambat. Kesimpulan tersebut terbatas pada kondisi penelitian dan periode pengamatan 8 HST. Supervised by Kamsia Dorliana Sitanggang and Badrul Ainy Dalimunthe. This study evaluated 40%, 50%, and 60% ethanol for surface sterilization of robusta coffee shoot explants. The experiment was conducted at the Tissue Culture Laboratory of Universitas Labuhanbatu from May to July 2026. Inoculation was performed on 1 July 2026 and cultures were observed until eight days after planting. A one-factor completely randomized design with three treatments and six replications was used, resulting in 18 experimental units. The percentages of contamination-free explants were 33.33%, 50.00%, and 50.00% for 40%, 50%, and 60% ethanol, respectively. Mean contamination onset was 7, 7, and 8 days after planting. Fungal contamination was dominant. Analysis of variance on data coded as 100 for contamination-free and 0 for contaminated produced F = 0.192 and p = 0.827. The 5% DMRT assigned the same letter to all treatments. The Fisher-Freeman Halton exact test also showed no significant association between ethanol concentration and culture status (p = 1.000). Descriptively, 60% ethanol showed the most favorable tendency because it achieved the joint-highest contamination free percentage and delayed contamination onset.
