Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh berbagai konsentrasi bahan sterilan terhadap keberhasilan sterilisasi eksplan tunas muda tanaman jambu Jamaika (Syzygium malaccense L.) secara in vitro, meliputi persentase eksplan steril dan kontaminasi, waktu munculnya kontaminasi, persentase browning, serta jenis kontaminasi. Penelitian menggunakan tiga perlakuan sterilisasi, yaitu alkohol 30% + Clorox 5%, alkohol 50% + Clorox 10%, dan alkohol 70% + Clorox 15%, dengan masing-masing perlakuan terdiri atas enam ulangan sehingga diperoleh 18 unit percobaan. Eksplan yang telah disterilisasi ditanam pada media Murashige and Skoog (MS) dan diinkubasi pada suhu ±25°C dengan pencahayaan selama 16 jam per hari. Pengamatan dilakukan terhadap persentase eksplan steril, kontaminasi, browning, waktu munculnya kontaminasi, dan jenis kontaminasi. Data dianalisis secara kuantitatif menggunakan persentase dan nilai rata-rata, kemudian dianalisis menggunakan analisis ragam (ANOVA) pada taraf 5%. Hasil penelitian menunjukkan bahwa secara deskriptif perlakuan alkohol 70% + Clorox 15% menghasilkan persentase eksplan steril tertinggi sebesar 83% dan kontaminasi terendah sebesar 17%, serta mampu menunda munculnya kontaminasi hingga rata-rata hari ke-5 setelah tanam. Namun, perlakuan tersebut menghasilkan persentase browning tertinggi sebesar 67%. Perlakuan alkohol 30% + Clorox 5% menghasilkan persentase browning terendah sebesar 0%, tetapi menghasilkan eksplan steril sebesar 17% dan kontaminasi sebesar 83%. Perlakuan alkohol 50% + Clorox 10% menghasilkan eksplan steril sebesar 50%, kontaminasi sebesar 50%, dan browning sebesar 33%. Hasil analisis ANOVA menunjukkan bahwa perlakuan sterilisasi tidak berpengaruh nyata terhadap persentase eksplan steril pada taraf 5% (Fhitung = 3,158 < Ftabel = 3,682; p = 0,0717 > 0,05). Berdasarkan hasil penelitian, kombinasi alkohol 70% + Clorox 15% merupakan perlakuan yang paling efektif secara deskriptif dalam menekan kontaminasi dan memperlambat munculnya kontaminasi. Namun, perlakuan tersebut belum dapat dinyatakan sebagai perlakuan paling optimal secara fisiologis karena menghasilkan browning tertinggi dan masih ditemukan kontaminasi bakteri. Kata kunci: Sterilisasi, Eksplan Tunas Muda, Jambu Jamaika, Syzygium Malaccense L., Kultur In Vitro ================================================================================================= This study aimed to determine the effect of various sterilization concentrations on the success of sterilizing young shoot explants of Jamaica apple (Syzygium malaccense L.) in vitro, including the percentage of sterile explants and contamination, the time of contamination occurrence, the percentage of browning, and the types of contamination. The study used three sterilization treatments, namely 30% alcohol + 5% Clorox, 50% alcohol + 10% Clorox, and 70% alcohol + 15% Clorox, with six replications for each treatment, resulting in 18 experimental units. The sterilized explants were cultured on Murashige and Skoog (MS) medium and incubated at approximately 25°C with 16 hours of light per day. Observations were conducted on the percentage of sterile explants, contamination, browning, time of contamination occurrence, and types of contamination. The data were analyzed quantitatively using percentages and mean values and were further analyzed using analysis of variance (ANOVA) at a 5% significance level. The results showed that descriptively, the 70% alcohol + 15% Clorox treatment produced the highest percentage of sterile explants at 83% and the lowest contamination at 17%, while delaying the occurrence of contamination until an average of the fifth day after planting. However, this treatment resulted in the highest browning percentage of 67%. The 30% alcohol + 5% Clorox treatment produced the lowest browning percentage of 0%, but resulted in only 17% sterile explants and 83% contamination. Meanwhile, the 50% alcohol + 10% Clorox treatment resulted in 50% sterile explants, 50% contamination, and 33% browning. The ANOVA results showed that the sterilization treatment did not have a significant effect on the percentage of sterile explants at the 5% significance level (Fcalculated = 3.158 < Ftable = 3.682; p = 0.0717 > 0.05). Based on the results, the combination of 70% alcohol + 15% Clorox was descriptively the most effective treatment for suppressing contamination and delaying its occurrence. However, this treatment could not be considered physiologically optimal because it produced the highest browning percentage and bacterial contamination was still observed.
