Abstract

Tanaman matoa (Pometia pinnata) merupakan salah satu tanaman yang memiliki potensi untuk dikembangkan melalui teknik kultur jaringan. Salah satu faktor penting dalam keberhasilan kultur jaringan adalah proses sterilisasi eksplan untuk mengurangi kontaminasi mikroorganisme. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi alkohol dan natrium hipoklorit (NaOCl) terhadap keberhasilan sterilisasi eksplan tanaman matoa secara in vitro serta menentukan perlakuan yang memberikan hasil terbaik. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Tanaman, Fakultas Sains dan Teknologi, Universitas Labuhanbatu. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan satu faktor yang terdiri atas tiga perlakuan dan enam ulangan. Perlakuan yang digunakan yaitu F1: alkohol 30% + Clorox 5%, F2: alkohol 50% + Clorox 10%, dan F3: alkohol 70% + Clorox 15%. Eksplan yang digunakan berupa tunas pucuk tanaman matoa dan ditanam pada media Murashige and Skoog (MS). Parameter yang diamati meliputi persentase eksplan steril, kontaminasi, waktu muncul kontaminasi, browning, serta jenis kontaminasi bakteri dan jamur. Data dianalisis menggunakan analisis ragam (ANOVA) pada taraf 5% dan dilanjutkan dengan uji BNT 5% apabila terdapat pengaruh nyata. Hasil penelitian menunjukkan bahwa secara deskriptif F3 memberikan persentase eksplan steril tertinggi sebesar 66,67%, diikuti F2 sebesar 50,00% dan F1 sebesar 16,67%. Persentase kontaminasi pada F1, F2 dan F3 masing-masing sebesar 83,33%, 50,00% dan 33,33%. Kontaminasi pada F1 dan F2 mulai muncul pada 3 HST, sedangkan F3 baru menunjukkan kontaminasi pada 6 HST. Pada parameter browning, F1 menghasilkan 0,00%, F2 33,33%, dan F3 66,67%. Hasil ANOVA menunjukkan bahwa perlakuan tidak berpengaruh nyata terhadap persentase eksplan steril dan kontaminasi pada taraf 5%, tetapi berpengaruh nyata terhadap browning pada taraf 5% dengan F hitung 3,7500 dan p-value 0,0478. Berdasarkan hasil penelitian, F3 (alkohol 70% + Clorox 15%) merupakan perlakuan yang paling baik secara deskriptif dalam menekan kontaminasi dan menghasilkan persentase eksplan steril tertinggi, tetapi penggunaan konsentrasi bahan sterilan yang lebih tinggi juga meningkatkan persentase browning eksplan. Kata kunci : Alkohol, Browning, Clorox, Eksplan, Kultur Jaringan, Matoa, Naocl, Sterilisasi. ==================================================================================== Matoa (Pometia pinnata) is a plant with considerable potential for propagation through tissue culture techniques. One of the important factors determining the success of tissue culture is explant sterilization to reduce microbial contamination. This study aimed to determine the effect of alcohol and sodium hypochlorite (NaOCl) combinations on the success of in vitro sterilization of matoa explants and to determine the treatment that provided the best results. The study was conducted at the Plant Tissue Culture Laboratory, Faculty of Science and Technology, Universitas Labuhanbatu. A Completely Randomized Design (CRD) with one factor consisting of three treatments and six replications was used. The treatments were F1: 30% alcohol + 5% Clorox, F2: 50% alcohol + 10% Clorox, and F3: 70% alcohol + 15% Clorox. The explants used were shoot tips of matoa plants cultured on Murashige and Skoog (MS) medium. The observed parameters included the percentage of sterile explants, contamination, time of contamination appearance, browning, and types of bacterial and fungal contamination. The data were analyzed using analysis of variance (ANOVA) at the 5% significance level, followed by the 5% Least Significant Difference (LSD) test when a significant effect was observed. The results showed that, descriptively, F3 produced the highest percentage of sterile explants at 66.67%, followed by F2 at 50.00% and F1 at 16.67%. The contamination percentages in F1, F2, and F3 were 83.33%, 50.00%, and 33.33%, respectively. Contamination first appeared on the third day after planting (3 DAP) in F1 and F2, while F3 showed contamination only on the sixth day (6 DAP). For the browning parameter, F1 showed 0.00%, F2 33.33%, and F3 66.67%. ANOVA results indicated that the treatments had no significant effect on the percentage of sterile explants and contamination at the 5% level, but significantly affected browning at the 5% level, with an F-value of 3.7500 and a p value of 0.0478. Based on the results, F3 (70% alcohol + 15% Clorox) was descriptively the best treatment for suppressing contamination and producing the highest percentage of sterile explants; however, the higher sterilant concentration also increased the percentage of browning in the explants.
